Hi Filiberto,
I would recommend purchasing the latest revision (2016) of ASHRAE110. This standard will include instructions on measurements for the type of fume hoods you have as well smoke visualization and measurement.
At our facilities, we perform ASHRAE 110 for the following: new installations, moves, and periodic (one every 5 years). Otherwise, we are performing yearly PMs (checking physical parts and airflow monitors) and face velocity measurements the same as prescribed in the ASHRAE 110.
To answer your questions specifically, you will perform measurements across the face (anemometer mounted on a movable stand, not handheld) across a grid pattern, taking the average 20 measurements.
